SPR is the most widely used technology for kinetic studies of label-free, real time biomolecular interactions. We present the ProteOn™ XPR36, an SPR optical biosensor offering parallel sample processing using a novel microfluidics system to achieve a 6 x 6 interaction array allowing simultaneous kinetic measurements of 36 interactions. We present two studies to illustrate the advantages of multi-spot kinetic studies in molecular recognition. The first involves antibody-antigen interactions. Interleukin 2 and a monoclonal antibody specific to interleukin 2 were characterized in one half-day experiment. Next, four interactions between IL2, IL4, IL6, and IL18 and monoclonal antibodies specific to these interleukins were characterized in one half-day experiment. The second set of experiments involves enzyme-inhibitor interactions: first, a single interaction between wild type TEM and BLIP-F142A was characterized in one half-day experiment. Next, five interactions between five different mutants of TEM and BLIP-F142A were characterized in one half-day experiment. For both studies, the second experiments were made possible because optimal experimental conditions could be determined from the first experiments due to ProteOn's novel parallel sample processing design.
