Analysis of intracellular materials at the single cell level presents opportunities for probing the heterogeneity of a cell population. Electrical lysis has been investigated as a rapid method for disruption of the cell membrane in the literature. In a typical single cell analysis experiment, electrical lysis provides the means for releasing the intracellular materials which are subsequently analyzed by assays such as electrophoresis. In this report, we studied the release of intracellular materials at the single cell level during electrical lysis. We examined the release of a small molecule, calcein (MW~600), and a 72 kDa protein kinase, Syk, tagged by enhanced green fluorescent protein (EGFP) from chicken B cells during electrical lysis at the single cell level. We studied the effects of the field intensity and field duration on the release of the two molecules. We found that calcein in general was released at lower field intensities and shorter durations than did SykEGFP. The field duration had more effect on the release of calcein than on that of SykEGFP. Taking advantage of the phenomena, we were able to deplete calcein from the cells before SykEGFP started to be release. Our results indicate that the release process during electrical lysis can vary significantly from one molecule to another. This needs to be taken into consideration when electrical lysis is performed for single cell analysis.
