As new methods for genomic engineering have been developed, a need for cost effective DNA sequencing has emerged. To improve the rate at which relevant genomic information can be sequenced, a technique for condensing genomic information has been developed. Under this method, targeted sites on the genome of E. coli were condensed into a single compact construct by overlap extension PCR using specially designed primers. A scalable kinetic model was developed and employed to optimize PCR parameters.
