First Report of PCR-RFLP Genotyping of Toxoplasma gondii from Monkey, Macaca mulatta : A BTUB Marker Allele Missing
Bsckground: The genetic variability among T.gondii (Types I, II and III ) isolatesd from different hosts. In the genetic, the rhesus mulatta(Macaca mulatta) is similar to the human. But at present , there were few studies about the genotyping of T.gondii from rhesus mulattas.
Method: In this study, a real-time PCR targeting the B1 gene of T.gondii was used to detect 38 positive samples from 40 rhesus mulattas’ blood samples. tissues. We genotyped 38 T.gondii DNA extracted from rhesus mulattas blood tissues. ,there are only 35 sampies strain typing was partly performed using 10 PCR-RFLP markers (SAG1, 5’-and 3’-SAG2, alt.SAG2,SAG3,BTUB,c22-8,c29-2,L358,PK1,Apico).
Result: Due to Toxoplasma gondii strains relatively rare in the blood, eight were successfully genotyped at three or more geneticloci, The most samples belong to T.gondii Genotype I, the 11th sample includes T.gondiiGenotype I in in 5’-and 3’-SAG2 markers but II in SAG3 marker, the 2th sample BTUB marker allele includes Genotype I in 5’-and 3’-SAG2 markers and u-3(our experimental team found a BTUB marker allele missing and name)in SAG3 marker.
Conclusions: To our knowledge ,this study fistly uses PCR-RFLP genotyped T.gondii strains from rhesus mulattas blood tissues and indicated that T.gondii I lineages is popular in rhesus mulattas and some new typical genotypes appear. This will facilitate the identification of T.gondii in population genetic studies.
