Complete Genome Analysis of Bacillus methylotrophicus CBMB205 Isolated from Rhizosphere Soil of Rice
Bacillus methylotrophicus, designated strain CBMB205, was isolated from the rhizosphere soil of field-grown rice. Cells were Gram-positive and rod-shaped. The 16S rRNA gene comparative analysis showed that strain CBMB205 form a clade with the species Bacillus amyloliquefaciens, B. vallismortis, B. subtilis, B. atrophaeus and B. mojavensis. CBMB205 was known for methanol-utilizing function, ACC deaminase activity, siderophore production and sulfur oxidation activity. And progressing study, CBMB205 has cps gene related cold stress, indole-3-glycerol phosphate synthase activity related auxin synthesis cycle, gene related heavy metal resistance and zeatin related gene as known for major cytokinin. So, it is considered to be useful for plant growth. However, genome sequencing of this strain was not performed yet. In this study, we investigated the genetic properties using a next generation sequencing (NGS) technology. The genomic DNA was isolated and sequenced by using PacBio platform with 259 coverage. Total 77,620 reads were produced, and these were assembled by PacBio SMRT Analysis 2.3.0. The genome of B. methylotrophicus CBMB205 had 3,929,745 base pairs circular chromosome with 46.50% GC contents and consists of 2,846 genes. It contained 27 rRNA and 86 tRNA sequence, 10% and 8% of the total genes involved amino acid transport and metabolism and transcription, respectively.
