Development of Species-Specific SCAR Markers in Eucalyptus
Of 59 primers tested in BSA, two (OPAD-01 and OPH-03) allowed differ two species. Primer OPAD-01 was a unique band to Eucalyptus saligna approximately 750 bp. Primer OPH-03 in approximately 700 bp was absence band in E. tereticornis, suggesting a negative marker. We opened the bulks for polymorphism confirmation and band selection for build SCAR markers. SCAR markers were validated on 30 individuals of species that marker was developed and 10 of other species (Eucalyptus grandis, E. tereticornis, E. brassiana, E. urophylla and E.saligna). Primer CAS was 90% efficiency in detection of E. saligna. Primer CHT showed no bands in E. tereticornisand band presence in other species at different percentages (20-60%). These SCAR primers can be used in the initial screening of Eucalyptus breeding programs and we encourage further studies with other Eucalyptus species, increasing primers number.
