Wheat (Triticum aestivum) is the most important cereal crops in the worldwide. However sequencing the wheat genome has been a challenge because of its huge and highly repetitive genome. The most accurate and efficient way to decode the large and complex genome is to sequence the minimum tilling path (MTP) BACs with the combination of next generation sequence (NGS) platform and BAC pooling strategy. In this study, we are sequencing 2312 MTP BACs derived from the short arm of Aegilops tauschii 3D chromosome. Briefly, 13 fragment libraries with 20-24 barcoded DNA pools were constructed, each BAC pool contains 8 MTP BACs. A GS FLX Titanium XL Plus run were performed for each library with sequence coverage of each BAC pool ranging from 15x to 43.1x, and average read length of 640bp. Eight 3kb pair end libraries of pooled 380 MTP BACs was constructed and sequenced 10 times coverage. The detailed sequence assembly and data analysis will present.
