Identification of Molecular Markers Associated to Puccinia melanocephala
The sugarcane programs have big challenges to keep and increase the annual production and one of the biggest challenges is to obtain genotypes resistant to pathogens. The Puccinia melanocephalais the fungus responsible for the brown rust, accounting for declines in productivity.
The identification of QTLs in bi-parental populations can aid in the identification of candidate genes for resistance and allow the marker-assisted selection in the future. Molecular markers such as target region amplification polymorphism (TRAP) that uses one primer derivated from expressed sequence tags (EST) and another one randomic are a powerful tool to support in locating these QTLs. In the present work there was developed (SUCEST database) and analyzed 16 TRAP markers that were analyzed at plant cane and ratoon cane, using a bi-parental cross between IACSP95-3018 clone and IACSP93-3046 variety, IAC breeding program. The individuals were phenotypically examined for resistance to rust, with scores of zero to nine.
Amplicons were obtained and visualized in 4300S DNA Analyzer. Were generated 466 markers that had segregation in a single dose (1:1 or 3:1). The single marker analyze showed twenty different significant markers, distribuited in thirty three associations, being eight of them significant in more than one period evaluated. Among the markers associated with plant cane and ratoon cane, some markers can assign as main, since they were significant at less than1%. These preliminary results show that TRAP have potential to generate markers to locate genome regions associated with brown rust resistance.
