Recombinant substitution lines (RSL) derived from single chromosome substitution lines of two spring varieties (Sandra/CP3B and Zlatka/CP3B) were used to create mapping population (near-isogenic lines; NIL´s) for fine mapping of new flowering time gene. This gene has been located as QTL on chromosome 3B of alternative wheat variety Èeská pøesívka and was designated as QFt.cri-3B.1. It causes change in wheat flowering time within range of several days. Segmental substitution lines were identified and developed from segregating progeny to fine map the gene. For better understanding of the way this gene influences plant growth, analyses of wheat apices development were also performed.

Novel Vrn-B1c allele has been identified in hexaploid spring wheat varieties using molecular approach (primer designing, sequencing etc.). Analysis of DNA sequence revealed both duplication and deletion within first intron of Vrn-B1 allele and multiplex PCR able to distinguish all four known Vrn-B1 alleles using one reaction was designed. Detailed investigation for a possible source of this novel allele was carried out. This work has been supported by the Czech Science Foundation (P501/10/1778).
