Capture-based target sequencing is a very effective strategy for deep scanning the region of interest in both human and non-human primate. Using this strategy, we have re-sequencing several candidate genes elucidated recently, including EPAS1 and EGLN1, which may responsible for human high altitude adaptation in number of Tibetan and Han Chinese individuals. Very interesting pattern of selected haplotype were observed. We also tried to use human design exon-capture array to recover coding region of non-human primate, including chimpanzees, crab-eating macaques and Japanese macaques. The strategy was proven to have good recovery rate and able to generate high quality variant call. Taken together, capture-based target sequencing still an attractive, cost-effective approach for the comparative analysis of human and non-human primate genomes.
