Although a great deal of rice proteomic research has been conducted, there are relatively few studies specifically addressing the rice grain and pollen proteome. Here, we performed shotgun proteomic analysis of waxy rice grain and quantitative shotgun proteomic analysis of rice pollen development to construct an in-depth proteome reference map, monitoring the expression patterns of the identified proteins, and to detect proteins that are expressed differentially during pollen development. Extracted proteins were separated in 1D-SDS-PAGE then the gel was sliced into seven. Chopped gels were ingel-digested. Peptides were subjected to mass spectrometry (Q-Exactive) ms/ms spectra were analyzed through Proteome Discoverer. We identified 1,069 non-redundant proteins with a wide range of molecular weights (from 5.2 kDa to 611 kDa) and pI values (from pH 2.9 to pH 12.6). Rice Anthers were harvested in 20 days before flowering, 10 days before flowering, and flowering day. The protein expression patterns were revealed by a quantitative shotgun proteoemic analysis. By merging all of the identified proteins in the three time points, total 4,509 anther proteome reference map was constructed. Proteins expression patterns were investigated by comparing the quantity of protein. The proteins expression patterns were identified in this study were clustered. Clustering analysis and Genome Ontology category enrichment analysis revealed that proteins involved in the metabolic process, gene expression, and translation were enriched through all stages of anther development.
