Artemisia frigida, is a Mongilian traditional medicinal plant and called ‘Agi’ by Mongolian. It belongs to the genus of Artemisia in composite family, distributing widely in Inner Mongolia Autonomous Region. This plant has the power of stanch, detumescence, and was well applied in the cure of bleeding, arthroncus, and so on. Few research reports are available on its molecular genetic basis. In the paper, one sequencing run of Artemisia frigida genomic DNA was carried out using Roche 454 GS FLX+ system. Simple sequence repeats (SSRs) were searched by MISA software. A total of 645,965 quality-filtered sequence reads were generated with the average read length of 598 bp, representing 387 Mb sequence data. Assembly of the nucleotide sequence reads was performed to obtain non-redundant contigs and singletons using the GS De Novo Assembler. In total, we got 28,129 contigs and 280,593 singletons. 12,428 SSRs motif were found by MISA software, which showed that average distribution frequency was 4.03%, and every 15.15 kb sequences could get one SSR in the Artemisia frigida genome. The number of dinucleotide and trinucleotide SSR were accounting for more than 80%. AC/TG, AT/TA and AAT/TTA were the main types of SSR repeat motifs in dinucleotide and trinucleotide SSR motifs. The repeat number of most of Artemisia frigida SSRs was from 5 to 10. The motif length of most of Artemisia frigida SSRs ranged from 12 bp to 30 bp. This research will be a powerful resource for the breeding and molecular biology studies of Artemisia frigida.
