Limited studies have been conducted of the diploid progenitor Aegilops speltoides, and very few genes of high molecular weight glutenin genes (HMW-GS) had been submitted. May be due to its questionable genome; the (SS) genome that is share among other Aegilops sp. or possibly BB genome contributor. One complete open reading frame (ORF) of the HMW-GS (1Sy18*) genes was cloned, sequenced and annotated comprehensively. Novel (JQ680980.1) gene and homolog putative protein (AFJ97497.1) was revealed. The qualitative protein patterns profile was explored using SDS-PAGE. Unique Cystine residues that have very important existence, and dependent on its location in the conserved N- and C- terminals of the HMW-GS proteins structure  were studied, with focusing on the specific number of repeats across the protein sequence which is associated with the type of protein (x and y). Novel discussion about the frequency rate and the importance of dipeptide motif (QQ) structure was displayed. The frequency of the independent motif as well as contributory frequency with other motifs was scored as the highest motif frequency via all the HMW-GS encoded proteins in the GeneBank database.
