The NCBI eukaryotic genome annotation pipeline uses RNA-Seq, cDNA/EST, and protein alignments to generate whole genome annotation with corresponding RefSeq RNA and protein sequence products.  A RefSeq copy of the submitted Bubalus bubalis UMD_CASPUR_WB_2.0 genome assembly (Assembly accession GCA_000471725.1) was annotated using a combination of water buffalo RNA-Seq reads from 30 tissues collected from male and female animals (Project PRJEB4351), plus RefSeq and GenBank cDNAs, ESTs, and proteins from water buffalo, cow and human.  Annotation results include >27,000 genes and pseudogenes and >41,000 annotated proteins of which roughly 2,000 are tracked as either end-partial or with a disrupted CDS.  The presentation will provide an overview of NCBI’s eukaryotic annotation pipeline, initial annotation results for the water buffalo genome, and comparison to other Bovinae.
