Fast, accurate, reliable genotyping has become an essential requirement for molecular breeding, crop improvement and population genomics studies in plant species. Plant geneticists and seed companies need rapid, cost effective methods to efficiently genotype large amounts of samples that are compatible with these high-throughput workflows.

Crude extraction methods generate DNA that is high in salt and other PCR inhibitors, making amplification from these samples challenging. This is particularly true for the current generation of qPCR genotyping mixes that use probes or high resolution melt (HRM) analysis, where clean, pure DNA is required for correct identification of the SNP.

Here we show results from a major multinational seed company that has developed a rapid, inexpensive extraction method from soy leaf and seed that can be used directly in genotyping assays. DNA extracted using this method was tested in TaqMan SNP genotyping and HRM assays using reagents from Roche, ABI and Bioline.

Only the SensiFAST Genotyping Kit and the SensiFAST HRM Kit from Bioline were able to amplify and correctly type all samples, providing a fast, accurate and cost-effective method to genotype large numbers of soy samples.
