The nonsense-mediated mRNA decay (NMD) pathway serves important functions on mRNA quality control in eukaryotic cells. It can specifically identify and degrade aberrant transcripts harbouring a premature translational termination codon (PTC), thereby prevent the production of truncated proteins which may be deleterious to cell. Porcine mu subunit of Glutathione-S-transferases gene (GSTM2) was found to contain a premature termination codons (PTCs) triggering NMD. GSTM2 mutation may be result in pre- or post-natal death in piglets. In order to investigate the GSTM2 gene function on embryonic development, we silenced GSTM2 to in vitro simulate mutated GSTM2 by RNA interference technology, and furthermore analyzed the network related to GSTM2 by deep sequencing.

We use next generation sequencing to analyze transcriptome between untreated ST cell and RNAi cell. 242 differentially expressed genes (q>0.8) were isolated, which significantly enriched in 156 pathways. Whereas, STAT1, SRC, DUSP family, CCL family, Integrin related genes, IL-8 etc participate in multiple pathways. Thus, in regard to GSTM2 functions, we have the following indications: 1) GSTM2 may regulate SRC to influence the expression of hormone which impacts on implantation of blastocysts; 2) GSTM2 may be directly or indirectly adjust some adhesion molecules of uterine luminal epithelial, such as OPN, MUC4, to influence the combination of matrix and placenta; 3) GSTM2 is likely to participate in the immune response of Maternal-Placental Interface; 4)GSTM2 may be directly or indirectly regulate STAT1 to impact on implantation of blastocysts. Furthermore, we verify differentially expressed genes involved in above pathways via q-PCR.
