To date the next-gen sequencing with massively parallel short-reads alone is not capable to de novo assemble the large and complex plant genomes. Moreover, very little of the daunting amount of next-gen sequence data has been translated into functional studies. The combination of the large DNA technology such as unbiased bacterial artificial chromosomes (unbiased BACs) and next-gen sequencing could revolutionize both genome sequencing and functional genomics. However, traditionally BAC libraries built by restriction partial digestion are inherently biased which cause some importance information missing and genomic gaps in sequence. To bridge these gaps, completely cover interested-target genomic regions, obtain the highest quality of genome sequencing assemblies, finish genomes, or functional applications, unbiased BAC technology is needed. We have successfully developed and continuously optimize novel technologies for unbiased large DNA isolation, purification, manipulation and Random Shear BAC cloning. Intact Genomics Inc. is the worldwide provider of the high-quality, unbiased Random Shear BAC libraries. We have also demonstrated broad applications of unbiased BAC libraries in genome studies and discoveries in plants and other species.
