The concentration of anthocyanins is one of the key factors determining cabbage leaf color. To develop molecular markers, to classify and selection of color-related genes, RNA Seq was carried out using 4 different kinds of cabbage: Purple cabbage(7S4-63, 7S4-51), Green cabbage(2409, 842), Cabbage accumulated anthocyanin at low temp.(337, 154), Cabbage no accumulated anthocyanin at low temp.(2437, 09WH-45). RNA Seq results identified the expression of genes related to anthocyanin synthesis through data analysis. MYB2, TT8, F3'H, DFR, ANS gene are highly expressed in the purple cabbage. Candidate SSR were isolated based on the RNA Seq data. Among them 45 SSR primer sets were confirmed by PCR, and nine primer sets were selected. Subject to confirmation by the cabbage F1 varieties one of them was able to distinguish between the green cabbage and purple cabbage. To analyze the genome structure of the 18 upstream gene and 15 structural gene related to the anthocyanin synthesis, we designed 33 upstream gene primer sets and 21 structural gene primer. F3'H, DFR, MYB2 genes was excavated a total of 24 SNP in the gene(11 Exon, 13 Intron) and 6 SSR(Intron). [This study was carried out by GSP.]
