P0627Identification of a New Microsatellite Marker Linked to Grain Filling Rate as an Indicator for Heat Tolerance Genes in a DH Wheat Population
Our main objective is to identify QTLs affecting quality and agronomic traits under heat stress. The doubled haploid (DH) population (Yecora Rojo X Ksu106) have been used in this study. The population was derived, from crosses between high-quality, heat-sensitive wheat cultivar (Yecora Rojo) and the adapted Saudi wheat line (Ksu106) having poor bread making quality, high heat tolerance and high yield potential, by microspores and anther culture methods. Field experiments for agronomic and quality traits evaluation have been conducted in two locations. Microsatellite (SSR) method have been carried out to determine the number of QTLs controlling the important agronomic and quality traits under heat stress and map them to their genomic positions. The analysis of variance indicated that the grain filling duration, grain filling rate and grain yield were significantly influenced by differences in heat treatment, wheat genotypes and their interaction. There were significant differences between Ksu106 and Yecora Rojo for all agronomic traits under heat treatments. Wide or continuous frequency distributions with transgressive segregants were observed, as one would expect for QTLs. Preliminary investigation have been conducted to optimize the SSR technique and to identify SSR markers linked to the grain filling rate as indicator for heat tolerance genes in the DH wheat lines (Ksu106 X Yecora Rojo).
