Aegilops tauschii is the donor of the D genome of hexaploid wheat (Triticum aestivum). Its large genome size and complexity impose a big challenge for sequencing and assembling the genome. Taking advantage of the available BAC-based physical map for the genome, MTP-based BAC pools were sequenced with Illumina’s MiSeq platform. A comprehensive approach by integrating BAC pools sequences, whole genome shotgun Illumina sequences, PacBio sequences, and BioNano maps was used to assemble the genome and ultimately construct pseudomolecules for each of the seven chromosomes. This work is part of the NSF-funded Project IOS-1238231 to generate a reference sequence for the genome of Ae. tauschii (http://aegilops.wheat.ucdavis.edu/ATGSP/).
