P1204A Rapid Method for Extraction and Purification of DNA from Dental Plaque, Tongue Scraping and Saliva Using IDeal Kit
DNA was extracted from samples using lysis buffer and a bead disruptor for 30 seconds followed by addition of an optimization buffer. After a brief centrifugation the isolated DNA was added directly to qPCR reaction and analyzed with universal bacterial 16S rDNA primers for gram-negative and gram-positive organisms. In addition, specific primers for the gram-negative Treponema denticola and Tannerella forsythia, and for the gram-positive Streptococcus mutans and Parvimonas micra were used.
The IDeal DNA extraction kit was found to be a simple and ultra-fast method suitable for extraction of DNA from saliva.
In addition, the IDeal DNA extraction was found to be ideal for isolation of sufficient quantities of DNA from small sample sizes and for isolation of sufficiently high quality DNA for use in multiple downstream molecular biology applications.
