Next-generation sequencing of RNA (RNA-Seq) is a rapidly evolving technology that can greatly benefit from development of simple, fast, and reproducible RNA-Seq sample prep procedures. We have developed a novel, Transposon-based method – TotalScript™ RNA-Seq – that produces stranded, cluster-ready RNA-Seq libraries in one day for sequencing on the Illumina® platform.  TotalScript RNA-Seq libraries can be made from ~5ng of high quality (RIN>7) total RNA without the need for poly(A) RNA enrichment or rRNA removal or from 100pg of poly(A)+ RNA. The libraries demonstrate high complexity, ~97-99% strandedness and low rRNA contamination.  The TotalScript procedure provides a simple, innovative and cost-effective solution to RNA-Seq sample prep.
