The Canadian Triticum Advancement through Genomics (CTAG) project is participating in the IWGSC through a commitment to sequence chromosome 1A.  The project is using the existing physical maps created for the 1AS and 1AL chromosome arms and the Illumina MiSeq platform for the sequencing of the minimal tiling path (MTP) developed for each arm.  The strategy is employing pair-end and mate-pair sequencing to enable robust assembly of individual BAC MTP clones.  A description of the steps to establish the optimal assembly strategy will be provided together with analysis of the assembled data.  This will include the trialing of a variety of genome assemblers, the impact of modifying key parameters on assembly efficiency and analysis of assembled data, including comparative analysis with genomic sequence data from other species, such as the recently described Triticum urartu (A genome) sequence.
