We present the improvements in MaSuRCA whole genome assembler that enabled us to produce the assembly of the biggest genome ever assembled -- the 32Gb Sugar Pine genome. We discuss the shortcomings of our techniques that we learned from the previous large tree genome project -- the Loblolly Pine assembly. We show that improving assembly techniques can result in vast improvement of the assembly quality when compared in the same initial data. The additional improvement mainly results from better pre-processing of the original sequencing data yielding much cleaner input to the actual assembly code. We also present preliminary results on impact of introducing data from novel sequencing technologies, such as single-molecule sequencing reads, on the assembly when combined with the existing Illumina data.
